Paraphyses IKI reaction in Scutellinia
Perz Piotr, 03-02-2007 23:10
Hi,

I have question about paraphyses IKI reaction in genus Scutellinia. Sometimes paraphyses stain in IKI green - it means they have carotenoids. Sometimes there is no IKI reaction - also paraphyses does not contains carotenoids.

How is the name for violet-lila reaction? Here paraphyses of Scutellinia pseudotrechispora (Schröt.) Le Gal in IKI

Bonjour,
Amicalement

I have actually observed the same in "Scutellinia sp." (see my paper on Lugol versus Melzer, mycotaxon 29: 428, 1987): when adding IKI (Lugol) to a water mount, the colour reaction of the paraphyses changes from greyish-violet to grey.olive or grey-brown. The violet reaction is seen at an iodine concentration of 0.05-0.1% while the olive appears at 0.1-0.2 % and higher. So the colour is concentration-dependent similar as in the hemiamyloid apical rings. But contrary to hemiamyloidity, the violet and olive are also seen when KOH-pretreated, and I believe also in Melzer.
