Hyalorbilia on a polyporus?
François Bartholomeeusen, 03-10-2018 22:13
Good evening forum members,

Can this be Hyalorbilia berberidis? With orbiliamycetes I feel worried because I then scan the limits of my microscope's capabilities.

Macroscopy: the ascomata have a diameter of 350 to 400 µm are young flattened spherical and later flattened dish-shaped, young light orange later light yellow-grey; scattered sessile on the substrate
Substrate: an old, decayed polyporus on a fallen branch of fagus sylvatica. In the vicinity grow some fruiting bodies of a slime mold namely Physarum spec. The ascomata grow both on the polyporus and on the heavily decayed wood.

Microscopie: (all dimensions of fresh material in water).

Asci : 9,43-13,43 x 1,16-1,4 µm; hyaline, cylindric-clavate, 8-track, inamyloid, rounded top covered with a gel layer, base short, firm and forked, arising from croziers.

Spore: 9,43-13,43 x 1.16-1.4 µm Q= 8,9; hyaline, narrowly fusoid, curved with sharp ends and vacuolar bodies in both poles;

Paraphyses : difficult to distinguish, same size as as asci and they do not exceed the asci, hyaline

Excipulum: hyaline with texture angularis/globulosa

Thank you for your help,

François Bartholomeeusen

Re : Hyalorbilia on a polyporus?
Dear Francois

splendid! And don't worry, my microsope is not better.

Could you please add the correct ascus size?

Trapping organs are mainly unknown in Hyalorbilia, and if they exist then they are small ellipsoid pegs on the hyphae.

I assume you refer to the four small apothecia on your macro. The large one is something else?

How many apothecia do you have? If you want you can send this for sequencing, e.g. to Guy Marson. It is the type of Hyalorbilia and we do not have a sequence of this species so far.

I would be glad to include this collection, would you mind sending me the data?

Re : Hyalorbilia on a polyporus?
Hello Zotto,

Thank you very much for your swift answer!
Hereby the correct measurements of the asci: 25,4-33,4 x 4,32-5,83 µm.
I found the frb's on 28 september 2018; België, Retie, Provenciaal Domein Prinsenpark;
GPS 51.225885 5044018 Lambert 197171 213098 IFBL C62124

I have re-examined that large apothecium in the first picture and it has a diameter of 443 µm and spores and asci are identical to the other apothecia. Several asci have a rounded base but luckily I also found asci with a forked base.

I have two small pieces of rotten wood covered with polyporus, each about 3 cm² in size. After some searching under the bino, I found a total of 19 frb's. Is that sufficient for sequencing? If necessary I can go back to look for new frb's, with some luck.

Many greetings,

François

Re : Hyalorbilia on a polyporus?
Thanks, indeed these are enough apothecia. But they are perhaps not that large:

When the large apo measures 440 µm, then the small are 150-200 µm.

GPS works as 51.225885 5.044018

I will ask Guy if he is able and willing to try this.

Re : Hyalorbilia on a polyporus?
Dear Francois

I have asked Guy and he said you may send the sample to him, but please before 17. October. His addresse is

Guy Marson
45b, rue de Bettembourg
L-5810 Hespérange,
Luxembourg

Important is that you keep the sample air-dried. Did you dry it immediately after study? When kept for some days in the fridge it may already get contaminated.

Re : Hyalorbilia on a polyporus?
Good evening Zotto,

I kept the sample wet until October 6. To be sure, I examined one of those smaller apothecia: diameter 305 µm and found the same features as the first sample.Unfortunately the woud was contaminated with Collembola and I found fewer frb's. So I air-dried it but now it is difficult to find frb's. Tomorrow I will go back looking for the substrate and hope to find new frb's. If I succeed I will send the material to Guy Marson.

Best regards

François

Re : Hyalorbilia on a polyporus?
Unfortunately, my attempt to find new fruiting bodies did not succeed. I will send the few dried ascomata by post to Guy Marson. Hopefully it will be enough for sequencing.

Best regards,

François

Re : Hyalorbilia on a polyporus?

Bien cordialement,
Guy

Re : Hyalorbilia on a polyporus?
Un grand merci Guy! Heureusement, mes prières ont été entendues parce que j'avais peu confiance en l'exsiccate.

Salutations,

François
